? 自然或人工选择法(如 Ts株)对人低毒或无毒的

? 优点:模拟自然感染,免疫效果好,局部 IgA。
? 理论上的缺点,实际工作中少见
?变异株可能回复到有毒力的野生株
?如果机体免疫缺陷,减毒株仍可能引起感染或并发症
?可能激活机体内其他病毒的感染
?减毒活苗可能引起持续感染
? 成功的范例,1961年我国应用脊灰疫苗,脊灰发
病率大幅度下降。 1980年天花病毒灭绝
遗传重组疫苗
? 将野毒株表面抗原基因与弱毒株其他基
因组合而获得减毒活病毒
? 适用于基因组分节段双链 RNA病毒,如
流感病毒、轮状病毒等
基因缺失活疫苗
? 利用 DNA重组技术,定向缺失病毒基因
组的某一区段,使病毒丧失毒力而保留
增殖能力和免疫原性
? 优点:疫苗株的遗传背景清楚,不易突
变回到野毒株
载体活疫苗
? 利用 DNA重组技术,将编码病毒抗原的基
因插入到另一无毒力的载体病毒,使之高
效表达
? 易建立多价疫苗
? 载体病毒常用的有痘苗病毒、腺病毒、
HSV-1
灭活疫菌( Killed vaccine)
? 灭活全病毒疫苗

灭活全病毒疫苗
? 物理或化学方法灭活病毒。适用烈性或易变

?无局部抗体,不激活 CTL,可激活 TDTH

? 优点
?不含病毒核酸,仅有能诱导中和抗体的衣壳

? 优点
?制备容易,可大量生产,易保存,副作用少

基因工程疫苗
? 利用 DNA重组技术制备的生物制品。如将
编码病毒特异抗原的基因用适当的载体将
此基因带入大肠杆菌或真核细胞,使之表

? 研发成本高,生产成本低,下游工程复杂
核酸疫苗
? 将病毒基因导入 DNA载体(通常是带有真
核表达调控基因的质粒或病毒),将重组
DNA导入机体,重组 DNA可以在细胞的转
录和翻译系统表达出目的蛋白
? 优点:同时刺激产生体液和细胞免疫,能
诱导 CTL活性,易生产,易保存
? 理论上缺点:安全性尚待考察
? 目前仅在实验研究中,尚未投放市场
独特型病毒疫苗( Idiotype)
? 基于 Jerne的免疫网络学说,内影像组抗体
中含有与抗原一致的表位序列,又称抗独
特型抗体疫苗
? 特别适合于
?不能培养或培养困难、产量极低的病原体
?直接用病病制备有潜在危险的,如 Retrovirus
?免疫原性弱,且不能用重组技术生产的抗原

TK- virus cannot grow in neural cells because

“Thymidine kinase” is a misnomer

抗病毒化学制剂
? 理论上病毒复制的任何环节都是抗病毒药
物发挥作用的 靶位置,但要区别病毒复制
与宿主细胞正常生理过程是很困难的。因
此大多数抗病毒药物的应用都有限制,甚
至对机体有毒性作用
? 抗病毒药的机制多是在药物进入宿主细胞
后受病毒的某种酶的作用形成具有抗病毒
活性的新的成份

?HIV的 晚期蛋白酶将病毒子代大分子蛋白裂
解为 病毒结构蛋白(形成成熟病毒体核心成
份)和病毒逆转录酶分子。 Saquinavir抑制
晚期蛋白酶的活性,因此不成获得成熟病毒
认壳蛋白和逆转录酶,但对宿主细胞蛋白酶
没影响
?与逆转录酶抑制剂联合使用可以减低血液中
HIV的含量,延长存活期,但对细胞内的
HIV作用差
